Language changes to Lab Methods and to Statistical Methods section for ADCC template. Initially noticed by Bhavesh during his review of PR 109 and send to lab for review. While we wait for the lab, we will leave these questions in a git issue and push the current version. ADCC template can be modified in the future.
In the "Lab Methods" section the current template language is: "Effector cells were PBMCs obtained from a HIV-seronegative donor with heterozygous for FcγR3A at position 158 (158F/V)." We are wondering if it should be changed to "Effector cells were PBMCs obtained from a HIV-seronegative donor with heterozygous for FcγR3A at position 158 (158F/V) or FcγR2A at position 131 (131H/R)."
The response call in the statistical methods section is currently described as: "A response is defined as positive if the peak baseline-subtracted % specific killing activity greater than or equal to 10% for either the 1:50 or 1:200 dilution." We are wondering if the word "peak" should be removed which would mean this sentence would be changed to: "A response is defined as positive if the baseline-subtracted % specific killing activity greater than or equal to 10% for either the 1:50 or 1:200 dilution." If you wouldn't mind sharing the scientific intuition behind your choice, please let us know as well. We were discussing this and thinking: if the prozone effect only impacts dilution 1:50, then the peak dilution would necessarily only be 1:50 or 1:200 (and otherwise it would be an error in the reading, as the readings should go down monotonically past 1:50 or 1:200). Therefore, the peak would be at either 1:50 or 1:200 and either sentence would be technically true. However, perhaps it's more clear to remove "peak". Let us know your preference.
